Descripción: Malonyl-CoA is formed by acetyl-CoA carboxylase-mediated carboxylation of acetyl-CoA. Fatty acid synthase catalyzes the NADPH-dependent condensation of malonyl-CoA and acetyl-CoA to produce palmitate. Proliferating human cancer cells upregulate this fatty acid synthesis pathway as a strategy for survival. High levels of malonyl-CoA, achieved through fatty acid synthase inhibition, proposed to contribute to cancer cell apoptosis.

Descripción: A number of tumor cells and cell lines have been observed to have highly upregulated expression and activity of fatty acid synthase (FAS). Inhibition of FAS by the irreversible inhibitor cerulenin leads to cytotoxicity and apoptosis in human cancer cell lines, an effect believed to be mediated by the accumulation of malonyl-coenzyme A in cells with an upregulated FAS pathway. Cerulenin also leads to profound weight loss and feeding inhibition in both high-fat diet wild type obese and leptin-deficient ob/ob mice.

Descripción: Acetyl-CoA carboxylase (ACC) is a complex multifunctional enzyme system. ACC is a biotin-containing enzyme which catalyses the carboxylation of acetyl-CoA to malonyl-CoA, the rate-limiting step in fatty acid synthesis. There are two ACC forms, alpha and beta, encoded by two different genes. ACC-alpha is highly enriched in lipogenic tissues. The enzyme is under long term control at the transcriptional and translational levels and under short term regulation by the phosphorylation/dephosphorylation of targeted serine residues and by allosteric transformation by citrate or palmitoyl-CoA. Multiple alternatively spliced transcript variants divergent in the 5' sequence and encoding distinct isoforms have been found for this gene.

Descripción: Coenzyme A (CoA) is an essential cofactor functioning as an acyl group carrier and carbonyl-activating group for the citric acid cycle and fatty acid metabolism. About 4% of cellular enzymes utilize CoA as a substrate. It is synthesized from pantothenic acid in a five-step process that requires ATP. The pantothenate kinase step of the CoA biosynthetic pathway is identified as a target for the development of antibacterial compounds.
