Descripción: APP Antibody: Accumulation of the amyloid-beta peptide (Abeta) in the cerebral cortex is a critical event in the pathogenesis of Alzheimer's disease. The beta-amyloid protein precursor (APP) is cleaved by one of two beta-secretases (BACE and BACE2), producing a soluble derivative of the protein and a membrane anchored 99-amino acid carboxy-terminal fragment (C99). The C99 fragment serves as substrate for gamma-secretase to generate the 4 kDa amyloid-beta peptide (Abeta), which is deposited in the Alzheimer's disease patient's brains. Recently, Death Receptor 6 (DR6) was found to interact with an amino-terminal fragment of the Beta-amyloid protein (N-APP) in neurons, activating a caspase 6-dependent apoptotic event leading to axonal degeneration and pruning during development, suggesting that these two proteins are involved in neural development and may possibly play a role in Alzheimer's disease.

Descripción: MYST1 (MYST histone acetyltransferase 1,MOF) belongs to the MYST family of histone acetyltransferases, which are employed in the cell to bring about transcriptional regulation. The MYST family includes MYST1, is named for the founding members MOZ, yeast YBF2 and SAS2, and TIP60. All members of this family contain a MYST region of about 240 amino acids with a canonical acetyl-CoA-binding site and a C2HC-type zinc finger motif. Most MYST proteins also have a chromodomain involved in protein- protein interactions and targeting transcriptional regulators to chromatin. Although MOF is expressed in both males and females, it associates with the X chromosome only in males. MOF contains a zinc-finger domain that is used to contact the globular part of the nucleosome and histone H4. The carboxy terminal domain of human MOF also has histone acetyltransferase activity directed against histones H3 and H2A, a characteristic shared with other MYST family histone.

The three subunits are encoded by distinct genes and belong to a family of membrane skeletal proteins involved in the assembly of spectrin-actin network in erythrocytes and at sites of cell-cell contact in epithelial tissues. Adducin, originally purified from human erythrocytes, was found to be a heterodimer of adducins alpha and beta. Polymorphisms resulting in amino acid substitutions in these two subunits have been associated with the regulation of blood pressure in an animal model of hypertension. Structurally, each subunit is comprised of two distinct domains. The amino-terminal region is protease resistant and globular in shape, while the carboxy-terminal region is protease sensitive. The latter contains multiple phosphorylation sites for protein kinase C, the binding site for calmodulin, and is required for association with spectrin and actin. Descripción: Small Ubiquitin-Related Modifier 1 (SUMO1) is an Ubiquitin-like protein that belongs to the ubiquitin family with SUMO subfamily. It is a family of small, related proteins that can be enzymatically attached to a target protein by a post-translational modification process termed sumoylation. SUMO1 functions in a manner similar to ubiquitin in that it is bound to target proteins as part of a post-translational modification system. This post-translational modification on lysine residues of proteins plays a crucial role in a number of cellular processes such as nuclear transport, DNA replication and repair, mitosis and signal transduction. SUMO1 is involved in a variety of cellular processes, such as nuclear transport, transcriptional regulation, apoptosis, and protein stability. SUMO1 is not active until the last four amino acids of the carboxy-terminus are cleaved off. Polymeric SUMO1 chains are also susceptible to polyubiquitination which functions as a signal for proteasomal degradation of modified proteins and may also regulate a network of genes involved in palate development.
