Descripción: The p28 subunit of interleukin 27 (IL-27), also known as interleukin 30 (IL-30), is a member of the interleukin 12 (IL-12) family of cytokines. p28 is a secreted polypeptide that associates with the Epstein-Barr virus induced gene 3 (EBI3) to form the IL-27 cytokine heterodimer complex. IL-27 functions as a proinflammatory cytokine that induces immunomodulatory effects in naïve CD4+ T cells, mast cells, and monocytes. p28 can also form a complex with cytokine-like factor 1 (CLF), that is secreted by dendritic cells, to regulate natural killer (NK) and T cell functions.

Descripción: TBX21 is a member of a phylogenetically conserved family of genes that share a common DNA-binding domain, the T-box. T-box genes encode transcription factors involved in the regulation of developmental processes. It is the human ortholog of mouse Tbx21/Tbet gene. Studies in mouse show that Tbx21 protein is a Th1 cell-specific transcription factor that controls the expression of the hallmark Th1 cytokine, interferon-gamma (IFNG). Expression of the human ortholog also correlates with IFNG expression in Th1 and natural killer cells, suggesting a role for this gene in initiating Th1 lineage development from naive Th precursor cells.

Descripción: The N418 monoclonal antibody specifically reacts with the integrin alpha x chain of the mouse CD11c, which is expressed on dendritic cells, CD4-/CD8+ intestinal intraepithelial lymphocytes (IEL) and some activated T lymphocytes. Low levels of CD11c were detected on mouse splenic natural killer cells and on the monocyte/macrophage lineage cells.CD11c expression is upregulated on IEL and T lymphocytes after activation. It binds to CD54, fibrinogen and iC3b and influences the leukocyte adhesive interactions. The N418 antibody binds to CD11c on splenic dendritic cells of the mouse in the T-dependent areas. It also contributes to the binding of iC3b.

Descripción: Function as an activating and costimulatory receptor involved in immunosurveillance upon binding to various cellular stress-inducible ligands displayed at the surface of autologous tumor cells and virus-infected cells. Provides both stimulatory and costimulatory innate immune responses on activated killer (NK) cells, leading to cytotoxic activity. Acts as a costimulatory receptor for T-cell receptor (TCR) in CD8(+) T-cell-mediated adaptive immune responses by amplifying T-cell activation. Stimulates perforin-mediated elimination of ligand-expressing tumor cells. Signaling involves calcium influx, culminating in the expression of TNF-alpha. Participates in NK cell-mediated bone marrow graft rejection. May play a regulatory role in differentiation and survival of NK cells. Binds to ligands belonging to various subfamilies of MHC class I-related glycoproteins.

Descripción: The RM4-5 monoclonal antibody specifically reacts with mouse CD4, also known as L3T4, a 55 kDa differentiation antigen expressed by the majority of thymocytes, subpopulations of mature T cells (like major histocompatibility complex class II-restricted T lymphocytes), a subset of natural killer T cells, and on pluripotent hematopoietic stem cells. CD4 binds to the major histocompatibility complex class II (MHC class II) and enhances T lymphocyte development and mature T cells functions. In T lymphocytes, CD4 binds to the cytoplasmic tail of enzyme tyrosine kinase (p56lck).Binding of RM4-5 is blocked by the anti-mouse CD4 clone GK1.5.

Descripción: GLT8D2 (glycosyltransferase 8 domain-containing protein 2), also known as GALA4A, is a 349 amino acid single-pass type II membrane protein. A member of the glycosyltransferase 8 family, GLT8D2 is encoded by a gene that maps to human chromosome 12q23.3. Encoding over 1,100 genes within 132 million base pairs, chromosome 12 makes up about 4.5% of the human genome. A number of skeletal deformities are linked to chromosome 12, including hypochondrogenesis, achondrogenesis, Noonan syndrome, Kniest dysplasia and trisomy 12p. Chromosome 12 is also home to a homeobox gene cluster, which encodes crucial transcription factors for morphogenesis, as well as the natural killer complex gene cluster, which encodes C-type lectin proteins that mediate the NK cell response to MHC I interaction.

Descripción: CD11b is a cell adhesion molecule that acts as a receptor for cell surface ligands such as intracellular adhesion molecules (ICAMs) or soluble ligands. Integrins are heterodimeric proteins that contain an a chain and b chain. Integrin αM combines with the Integrin β2 to form a leukocyte-specific integrin referred to as macrophage receptor 1 (Mac-1), or inactivated-C3b (iC3b) receptor 3 (CR3). Integrin αM/β2 is important in the adherence of neutrophils and monocytes to stimulated endothelium, and also in the phagocytosis of complement coated particles. The protein CD11b has been implicated in the various adhesion-related interactions of cells such as monocytes, macrophages, natural killer (NK) cells, and granulocytes. It is part of a heterodimer that consists of CD11b and CD18. It also modulates the uptake of complement-coated particles within the cell. It is commonly used as a microglial marker in tissues derived from the nervous system.

Descripción: Recognizes a protein of 40 kDa, identified as CD7, a member of the immunoglobulin gene superfamily. Its N-terminal amino acids 1-107 are highly homologous to Ig kappa-L chains whereas the carboxyl-terminal region of the extracellular domain is proline-rich and has been postulated to form a stalk from which the Ig domain projects. CD7 is expressed on the majority of immature and mature T-lymphocytes, and T cell leukemia. It is also found on natural killer cells, a small subpopulation of normal B cells and on malignant B cells. Cross-linking surface CD7 positively modulates T cell and NK cell activity as measured by calcium fluxes, expression of adhesion molecules, cytokine secretion and proliferation. CD7 associates directly with phosphoinositol 3'-kinase. CD7 ligation induces production of D-3 phosphoinositides and tyrosine phosphorylation.

Descripción: Interleukin 21 (IL-21) is a member of the common-gamma chain family of cytokines that are involved in immunoregulation. IL-21 is normally expressed by activated CD4+ T cells. The IL-21 receptor (IL-21R) activates the JAK/STAT signaling pathway and is expressed on T, B, and natural killer (NK) cells. Within the B cell lineage, IL-21 is a switch factor regulating IgG1 and IgG3 antibody production. IL-21 also cooperates with interleukin 4 (IL-4) for the production of multiple antibody classes in B cells. IL-21 has pleiotropic effects on the proliferation, differentiation, and effector functions of B, T, NK, and dendritic cells.
