Descripción: Ephrin-B1, also named EFL-3, ELK ligand, EPH-related receptor tyrosine kinase ligand 2, is a single-pass type I membrane protein. It contains 1 ephrin RBD (ephrin receptor-binding) domain and belongs to the ephrin family. Ephrins are divided into the ephrin-A (EFNA) class, which are anchored to the membrane by a glycosylphosphatidylinositol linkage, and the ephrin-B (EFNB) class, which are transmembrane proteins. All ephrins share a conserved extracellular sequence, which most likely corresponds to the receptor-binding domain. Ephrin-B1 has been shown to bind EphA3, EphB1, EphB2, EphB3, and EphB4. The extracellular domains of human and mouse ephrin-B1 share 94% amino acid identity.

Descripción: Epidermal growth factor (EGF) has a profound effect on the differentiation of specific cells in vivo and is a potent mitogenic factor for a variety of cultured cells. The EGF precursor is believed to exist as a membrane-bound molecule which is proteolytically cleaved to generate the 53-amino acid peptide hormone that stimulates cells to divide. EGF exerts its actions by binding to the EGFR, a 170 kDa protein. Epidermal growth factor is also a key growth factor regulating cell survival. Through its binding to cell surface receptors, EGF activates an extensive network of signal transduction pathways that include activation of the PI3K/AKT, RAS/ERK and JAK/STAT pathways

Descripción: The S100 family of proteins are found in vertebrates and are characterised by two calcium-binding sites that have a helix-loop-helix conformation. The family of 24 is divided into three subgroups based on function: those with intracellular, intracellular plus extracellular, and extracellular regulatory effects. The calcium-binding protein S100A8, also called MRP8 or Myeloid-related protein 8, and Calgranulin A, forms a heterodimer with S100A9, called Calprotectin. The heterodimer is expressed by granulocytes, monocytes, and early differentiation states of macrophages, and is known to be an important pro-inflammatory mediator in acute and chronic inflammation. S100A8 and 9 are DAMPs, or Damage-associated molecular pattern molecules, and are able to activate receptors of the innate immune system, one being TLR4.

Descripción: Bid Antibody: Apoptosis plays a major role in normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in a variety of diseases such as cancer. The Bcl-2 family of proteins is comprised of critical regulators of apoptosis that can be divided into two classes: those that inhibit apoptosis and those that promote cell death. Bid, a pro-apoptotic Bcl-2 family member, is cleaved by caspase-8 in response to apoptotic signals, exposing the Bcl-2 homology 3 (BH3) domain which is normally buried in the full-length protein. The cleaved complex is myris-toylated and translocated to the mitochondrial membrane where it may induce mitochondrial Bax and Bak to oligomerize.

Descripción: Crystallins are the major proteins of the vertebrate eye lens, where they maintain the transparency and refractive index of the lens. Crystallins are divided into a, b, and g families, and the b- and g-crystallins also comprise a superfamily. Crystallins usually contain seven distinctive protein regions, including four homologous motifs, a connecting peptide, and N- and C-terminal extensions. b-crystallins constitute the major lens structural proteins. They associate into dimers, tetramers, and higher order aggregates. The b-crystallin subfamily is composed of several gene products, including bA1-, bA2-, bA3-, bA4-, bB1-, bB2- and bB3-crystallin. The bA1- and bA3-crystallin proteins are encoded by a single mRNA. They differ by only 17 amino acids, and bA1-crystallin is generated by use of an alternate translation initiation site. The genes for bA4-, bB1-, bB2- and bB3-crystallin are clustered on human chromosome 22q11, while the genes for bA3/A1- and bA2-crystallin map to human chromosomes 17q11 and 2q34, respectively.

Descripción: Bcl-xL Antibody: Apoptosis plays a major role in normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in a variety of diseases such as cancer. Bcl-xL is a member of the Bcl-2 family of proteins that are critical regulators of apoptosis. These can be divided into two classes: those that inhibit apoptosis and those that promote cell death. Bcl-xL is an anti-apoptotic mitochondrial protein related to Bcl-w and the major transcript of the bcl-x gene. Its high expression in tumors is correlated with advanced disease and poor prognosis. Bcl-xL expression level increases in response to several stimuli such as ionizing radiation and treatment with chemotherapeutic agents.

Descripción: OCLN Antibody: Tight junctions act as a semi-permeable barrier to the transport of ions, solutes, and water and are considered to function as a fence that divides apical and basolateral domains of plasma membranes. Tight junctions coordinate a variety of signaling and trafficking molecules regulating cell differentiation, proliferation, and polarity and contain a number of junctional proteins including Occludin, Claudins, junctional adhesion molecules (JAMs), as well as multiple scaffold proteins. Occludin, the first identified component of tight junction strands, is thought function as a signal transmitter in multiple signaling pathways and can associate with multiple kinases and phosphatases such as PI3-kinase and protein phosphatases 1 and 2A. At least two isoforms of OCLN are known to exist.

Descripción: In eukaryotic systems, initiation of transcription from protein-coding genes is a complex process requiring RNA polymerase II and broad families of auxiliary transcription factors. Such factors can be divided into two major functional classes: the basal factors that are required for transcription of all Pol II genes, including TFIIA, TFIIB, TFIID, TFIIE, TFIIF and TFIIH; and sequence-specific factors that regulate gene expression. The basal transcription factors and Pol II form a specific multiprotein complex near the transcription start site by interacting with core promotor elements such as the TATA box generally located 25-30 base pairs upstream of the transcription start site. Binding of TFIID to the TATA element initiates assembly of the other factors into a pre-initiation complex. The TATA-binding subunit of TFIID (designated TFIIDt or TBP) from higher eukaryotes contains a highly conserved 180 amino acid C-terminal domain.

Descripción: PCDHGB5 is a member of the protocadherin gamma gene cluster, one of three related clusters tandemly linked on chromosome five. These gene clusters have an immunoglobulin-like organization, suggesting that a novel mechanism may be involved in their regulation and expression. The gamma gene cluster includes 22 genes divided into 3 subfamilies. Subfamily A contains 12 genes, subfamily B contains 7 genes and 2 pseudogenes, and the more distantly related subfamily C contains 3 genes. The tandem array of 22 large, variable region exons are followed by a constant region, containing 3 exons shared by all genes in the cluster. Each variable region exon encodes the extracellular region, which includes 6 cadherin ectodomains and a transmembrane region. The constant region exons encode the common cytoplasmic region. These neural cadherin-like cell adhesion proteins most likely play a critical role in the establishment and function of specific cell-cell connections in the brain. Alternative splicing has been described for the gamma cluster genes.

Descripción: Glutamate receptors mediate most excitatory neurotransmissions in the brain and play an important role in neural plasticity, neural development and neurodegeneration. Ionotropic glutamate receptors are divided into two categories, namely NMDA receptors and kainate/AMPA receptors, both of which contain glutamate-gated, cation-specific ion channels. Kainate/AMPA receptors consist of seven structurally related subunits, designated GluR-1 to -7, and are primarily responsible for fast excitatory neurotransmissions carried out by glutamate. GluR-delta 1 (Glutamate receptor delta-1 subunit), also known as GRID1, is a multi-pass membrane protein that belongs to the kainate/AMPA receptor family and is expressed primarily in the brain. Localized to the cell junction and the postsynaptic cell membrane, GluR-delta 1 functions as a glutamate receptor that regulates synaptic transmissions in the central nervous system (CNS) and is thought to play an important role in synaptic plasticity. Defects in the gene encoding GluR-delta 1 are associated with schizophrenia, a chronic and severe brain disorder.

Descripción: Microtubules, the primary component of the cytoskeletal network, interact with proteins called microtubule-associated proteins (MAPs).MAP9 is a microtubule-associated protein required for spindle function, mitotic progression, and cytokinesis. The microtubule-associated proteins can be divided into two groups, structural and dynamic. The MAP proteins function to stimulate tubulin assembly, enhance microtubule stability, influence the spatial distribution of microtubules within cells and utilize microtubule polarity to translocate cellular components. MAP-9 (microtubule-associated protein 9), also known as ASAP, is a 647 amino acid cytoplasmic protein that is constitutively expressed during the cell cycle. MAP-9 localizes to microtubules in interphase, associates with the mitotic spindle during mitosis and localizes to the central body during cytokinesis. Involved in organization of the bipolar mitotic spindle, MAP-9 is required for bipolar spindle assembly, mitosis progression and cytokinesis. MAP-9 may be involved in stabilizing interphase microtubules. Two isoforms of MAP-9 are produced due to alternative splicing events.
