Descripción: Lon Protease, is a member of the Lon protease family. They are found in archaea, bacteria and eukaryotes. Lon protease is ATP-dependent serine protease that mediates the selective degradation of mutant and abnormal proteins as well as certain short-lived regulatory proteins, including some antitoxins. ARFs are important in eukaryotic vesicular trafficking pathways and they play an essential role in the activation of phospholipase D (PC-PLD). ARL15 (ADP-ribosylation factor-like 15), is a member of the ARF family of the Ras superfamily of small GTPases that are known to be involved in multiple regulatory pathways altered in human carcinogenesis. Other members of this protein family include ARL11, thought to be a tumor suppressor and may play a role in the regulation of apoptosis, ARL2 which plays an important role in microtubule dynamics and cell cycle progression, and ARL4, which may play a role in neurogenesis during embryonic development. Descripción: EHEC is capable of inducing life threatening illnesses, particularly in people with immune deficiency, young children and the elderly. Although the most common cause of EHEC is <i>E. coli</i> O157, other serotypes, such as O26, O45, O103, O111, O121 and O145, are also relatively frequent causes of infection. Typical symptoms are severe abdominal pain and diarrhoea, which is initially watery but often becomes bloody. <i>E. coli</i> are gram-negative, non sporulating, facultative anaerobic bacilli. Amongst the pathogenic <i>E. coli</i>, Verotoxin-forming strains (STEC or VTEC) have gained importance in recent years. The group of enterohaemorrhagic <i>E. coli</i> (EHEC) with its highly pathogenic serovar O157:H7 strain is particularly interesting in this respect. The main sources of infection are contaminated, raw or insufficiently heated foods of animal origin, such as meat and dairy products. The reservoir for EHEC is the gut of ruminants. Microorganisms can enter food during the processing of meat and dairy products if hygienic conditions are inadequate. The drastic increase in the incidence of food contamination caused by <i>E. coli</i> O157 demands reliable and rapid methods of detection. Innovative, rapid tests make screening for pathogenic organisms easier. Complete test sets for the detection of pathogens and toxins. Brefeldin A is a fungal metabolite which is a macrocyclic lactone exhibiting a wide range of antibiotic activity. Produced by Penicillium brefeldianum. Blocks binding of the cytosolic coat protein b-COP and ARF to Golgi membranes mediated by protein G. Also blocks protein transportation into post-Golgi compartments. It activates the sphingomyelin cycle. Brefeldin A mediated apoptosis has been observed in human tumor cells.
Brefeldin A reversibly inhibits the intracellular translocation of proteins in eukaryotes, e.g., during transport of proteins to the cell surface for secretion or expression. It has been reported to block the response of cultured cells to cholera toxin. In HepG2 cells, BFA induces two blocks in the secretory pathway; one at the level of the endoplasmic reticulum-Golgi juncture and the other in the trans-Golgi network. Brefeldin A is used in the studies of Brefeldin A-inhibited Guanine Nucleotide-exchange Protein, BIG2, Regulates the Constitutive Release of TNFR1 Exosome-like Vesicles.
