Because of the relatively low water solubility of the AM ester, Pluronic F-127, a mild detergent, is often used as a dispersing agent to facilitate cell loading. Indo-1-AM Ester itself does not bind Ca2+ , but it is readily hydrolyzed to indo-1 by endogenous esterases once it enters cells.

Descripción: By immunohistochemistry, this antibodyspecifically recognizes a protein in melanocytes and melanomas. This MAb reacts with junctional and blue nevus cells and variably with fetal and neonatal melanocytes. Intradermal nevi, normal adult melanocytes, and non-melanocytic cells are negative. It does not stain tumor cells of epithelial, lymphoid, glial, or mesenchymal origin. Metastatic amelanotic melanoma can often be confused with a variety of poorly differentiated carcinomas, large cell lymphomas, and sarcomas using H & E stains alone. It is also difficult to differentiate melanoma from spindle cell carcinomas and various types of mesenchymal neoplasms. This MAb stains fetal and neonatal melanocytes, junctional and blue nevus cells, and malignant melanoma. This MAb also stains Angiomyolipoma (PEComa).

Descripción: Recognizes a 53 kDa protein, identified as the Ornithine Decarboxylase (ODC-1). ODC is the initial and rate-limiting enzyme in the biosynthetic pathway of polyamines and is involved in the conversion of ornithine to putrescine. The biological activity of ODC-1 is rapidly induced in response to virtually all agents known to promote cell proliferation including hormones, drugs, growth factors, mitogens, and tumor promoters. Reportedly, ODC mRNA levels are elevated in lung carcinomas as well as in colon adenomas and carcinomas. ODC activity in colorectal carcinomas is greater than those in adenomas and normal mucosa.

CD22 expression is restricted to normal and neoplastic B cells and is absent from other haemopoietic cell types. In B-cell ontogeny, CD22 is first expressed in the cytoplasm of pro-B and pre-B cells, and on the surface as B cells mature to become IgD . It is not expressed by plasma cells, CD22 is found highly expressed in follicular mantle and marginal zone B-cells, and while germinal center B-cells are relatively weak. CD22 is a member of the immunoglobulin superfamily and serves as an adhesion receptor for sialic acid-bearing ligands expressed on erythrocytes and all leukocyte classes. It also associates with tyrosine kinases and play a role in signal transduction and B-cell activation.

Descripción: Recognizes an integral membrane glycoprotein of 150 kDa, identified as CD13 (also known as aminopeptidase-N). The antibody recognizes an extracellular epitope. The CD13 antigen is present on most cells of myeloid origin including granulocytes, monocytes, mast cells, and GM-progenitor cells. It is also expressed by the majority of AML, CML in myeloid blast crisis, and in a smaller fraction of lymphoid leukemias. CD13 is absent from normal lymphocytes, platelets and erythrocytes. CD13 is also present on fibroblasts; endothelial cells, epithelial cells from renal proximal tubules and intestinal brush border, bone marrow stromal cells, osteoclasts, and cells lining bile duct canaliculi. CD13 is identical to aminopeptidase N (APN), a prominent membrane-bound metalloprotease present on the surface of intestinal brush border and renal tubules. CD13 plays a role in metabolism of biologically active peptides, in phagocytosis, and in bactericidal/tumoricidal activities. It also serves as a receptor for human coronaviruses (HCV). The lineage-restricted pattern of expression of CD13 within the hemopoietic compartment suggests that it may be important in myeloid cell differentiation.

Descripción: CD45R, also designated CD45 and PTPRC, has been identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. CD45RB is expressed on mature B-lymphocytes and the majority of lymphomas and leukemias of B-cell origin.

Descripción: Recognizes a protein of 55 kDa, identified as CD25. It is expressed on activated T- and B-cells and activated monocytes/macrophages. With respect to lymphomas, CD25 is present on malignant cells of Hodgkin's disease, HTLV-1 adult T-cell leukemia, cutaneous T-cell lymphoma, and hair cell leukemia. Increased levels of soluble CD25 are observed in the leukemias/lymphomas and inflammatory/ autoimmune diseases. CD25 molecule alone appears to function as a low affinity receptor and associates with CD122 (IL-2R chain, p75) and CD132 (common chain) to form the high affinity IL-2 receptor complex. CD25 antibodies detect three epitope regions, A, B and C. This MAb recognizes the epitope B, which is located at residue 3-104 of CD25 and doe not block IL-2 binding to CD25.
