Descripción: Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is well known as one of the key enzymes involved in glycolysis. It catalyzes an important energy-yielding step in carbohydrate metabolism, the reversible oxidative phosphorylation of glyceraldehyde-3-phosphate in the presence of inorganic phosphate and nicotinamide adenine dinucleotide (NAD). The enzyme exists as a tetramer of identical chains. Besides its functioning as a glycolytic enzyme in cytoplasm, recent evidence suggest that mammalian GAPDH is also involved in a great number of intracellular proceses such as membrane fusion, microtubule bundling, phosphotransferase activity, nuclear RNA export, DNA replication, and DNA repair. During the last decade a lot of findings appeared concerning the role of GAPDH in different pathologies including prostate cancer progression, programmed neuronal cell death, age- related neuronal diseases, such as Alzheimer’s and Huntington’s disease.

NMNAT plays a key role in NAD(+) biosynthesis, catalysing the condensation of nicotinamide mononucleotide and ATP, and yielding NAD(+) and pyrophosphate. NMNAT appears to be a substrate of nuclear kinases and contains at least three potential phosphorylation sites. The interaction of NMNAT with nuclear proteins is likely to be modulated by phosphorylation. NMNAT is widely expressed with highest levels in skeletal muscle, heart, liver and kidney.

GPDH-C plays a critical role in carbohydrate and lipid metabolism by catalyzing the reversible conversion of Dihydroxyacetone Phosphate (DHAP) and reducing Nicotine Adenine Dinucleotide (NADH) to Glycerol-3-Phosphate (G3P) and NAD+. GPDH-C is inhibited by zinc ions and sulfate. Mutations in this gene are a cause of transient infantile hypertriglyceridemia. GPDH-C is unlike Glyceraldehyde 3-Phosphate Dehydrogenase (GAPDH); they have different substrates.

Two forms of Nampt exist, an intracellular form (iNampt) and an extracellular form (eNampt). While the function of iNampt as an essential and rate-limiting NAD+ biosynthetic enzyme is well established, the physiological role of eNampt is still a matter of debate. Nampt has various functions, including the promotion of vascular smooth muscle cell maturation and inhibition of neutrophil apoptosis. It activates insulin receptor and has insulin-mimetic effects, lowering blood glucose and improving insulin sensitivity. The protein is highly expressed in visceral fat and serum levels of the protein correlate with obesity.
