Descripción: The RING-type zinc finger motif is present in a number of viral and eukaryotic proteins and is made of a conserved cysteine-rich domain that is able to bind two zinc atoms. Proteins that contain this conserved domain are generally involved in the ubiquitination pathway of protein degradation. MARCH8 (membrane-associated ring finger (C3HC4) 8), also known as MIR or RNF178 (RING finger protein 178), is a 291 amino acid multi-pass membrane protein that localises to vesicle membranes and contains one RING-CH-type zinc finger. Expressed in a variety of tissues, including immature dendritic cells, MARCH8 functions as an E3 ubiquitin ligase that is thought to regulate immune responses by promoting the ubiquitination and subsequent degradation of target proteins, such as B7-2 and CD71.

Descripción: DISP2 is the second of three known homologs of the D.melanogaster protein Dispatched. It is a multitransmembrane protein containing two PTCH/DISP domains and is thought to be involved in the release of lipid-anchored Hedgehog from producing cells. Hedgehog is a major player in signaling pathways during embryogenesis, tissue regeneration, and carcinogenesis and the DISP proteins have been implicated in these pathways. Recently, it has been shown that DISP2 is translationally regulated by the microRNA miR-214 in zebrafish. Expression of this miRNA decreased DISP2 promoter activity in vitro and its overexpression in zebrafish resulted in a phenotype identical to that observed by DISP2 mutants. At least two isoforms of DISP2 are known to exist.

Descripción: Sequence-specific RNA-binding protein that acts as a post-transcriptional repressor by binding the 3'-UTR of mRNA targets. Binds to an RNA consensus sequence, the Pumilio Response Element (PRE), 5'-UGUANAUA-3', that is related to the Nanos Response Element (NRE) (PubMed:21572425, PubMed:18328718, PubMed:21653694, PubMed:21397187). Mediates post-transcriptional repression of transcripts via different mechanisms: acts via direct recruitment of the CCR4-POP2-NOT deadenylase leading to translational inhibition and mRNA degradation (PubMed:22955276). Also mediates deadenylation-independent repression by promoting accessibility of miRNAs (PubMed:18776931, PubMed:20818387, PubMed:20860814, PubMed:22345517). Following growth factor stimulation, phosphorylated and binds to the 3'-UTR of CDKN1B/p27 mRNA, inducing a local conformational change that exposes miRNA-binding sites, promoting association of miR-221 and miR-222, efficient suppression of CDKN1B/p27 expression, and rapid entry to the cell cycle (PubMed:20818387). Acts as a post-transcriptional repressor of E2F3 mRNAs by binding to its 3'-UTR and facilitating miRNA regulation (PubMed:22345517). Involved in neuronal functions by regulating ATXN1 mRNA levels: acts by binding to the 3'-UTR of ATXN1 transcripts, leading to their down-regulation independently of the miRNA machinery (PubMed:25768905). Plays a role in cytoplasmic sensing of viral infection (PubMed:25340845). In testis, acts as a post-transcriptional regulator of spermatogenesis by binding to the 3'-UTR of mRNAs coding for regulators of p53/TP53. Involved in embryonic stem cell renewal by facilitating the exit from the ground state: acts by targeting mRNAs coding for naive pluripotency transcription factors and accelerates their down-regulation at the onset of differentiation (By similarity).

Descripción: MARCH8 Antibody: MARCH8 (c-MIR) is a novel E3 ubiquitin ligase designated as the modulator of immune recognition (MIR) family, whose catalytic domain is a variant RING domain (RING-CH domain). MARCH8 was found as a functional and structural homolog of KSHV MIR1 and MIR2. MARCH8 targets B7-2 to lysosomal degradation and down-regulates the B7-2 surface expression through ubiquitination of its cytoplasmic tail. Furthermore, MARCH8 has been shown to down-regulate the expression of transferrin receptor and Fas, an important molecule for the induction of apoptosis. MARCH8 is the first example of an E3 ubiquitin ligase that is capable of inhibiting MHC II expression. Recent findings suggest that MARCH8 may regulate immune responses by promoting ubiquitination of MHC-II and CD86, leading to their subsequent endocytosis and lysosomal degradation.

Descripción: Leukocyte immunoglobulin-like receptor subfamily B member 2 (LILRB2) is also known as CD85 antigen-like family member D (CD85d), Immunoglobulin-like transcript 4 (ILT-4), Monocyte / macrophage immunoglobulin-like receptor 10 (MIR-10), which is a member of the the subfamily B class of LIR receptors. LILRB2 is receptor for class I MHC antigens. LILRB2 recognizes a broad spectrum of HLA-A, HLA-B, HLA-C and HLA-G alleles. LILRB2 competes with CD8A for binding to class I MHC antigens. LILRB2 / CD85d inhibits FCGR1A-mediated phosphorylation of cellular proteins and mobilization of intracellular calcium ions.
