Descripción: P450 enzymes constitute a family of monooxygenase enzymes that are involved in the metabolism of a wide array of endogenous and xenobiotic compounds (1). Several P450 enzymes have been classified by sequence similarities as members of the CYP1A and CYP2A subfamilies (2). NADPH cytochrome P450 reductase is a microsomal enzyme responsible for the transfer of electrons from NADPH to cytochrome P450 enzymes during the P450 catalytic cycle (3,4). NADPH cytochrome P450 reductase is localized to the endoplasmic reticulum where it is also able to transfer electrons to heme oxygenase and cytochrome b5 (5,6). NADPH cytochrome P450 reductase is structurally related to two separate flavoprotein families, ferredoxin nucleotide reductase (FNR) and flavodoxin (7). Electron transfer of NADPH cytochrome P450 reductase requires the binding of two flavin cofactors, FAD and FMN, to the FNR and flavodoxin domains, respectively (8).

Descripción: HCV is classified into the genus Hepacivirus of the family Flaviviridae. Like all the members of the family, HCV is an enveloped, single-stranded, positive-sense RNA virus. Its genome (about 9600 nt) is flanked at both termini by conserved, highly structured non-translated regions (NTRs) and encodes a polyprotein precursor (about 3000 aa), which is proteolytically processed by host and viral proteases to produce the structural (core, E1, E2 and p7) and non-structural (NS2, NS3, NS4A, NS4B, NS5A and NS5B) proteins of the virus. Recently, an additional protein has been identified, whose function remains unknown. NS5A is a ~56 kDa pleiotropic protein with key roles in both viral RNA replication and modulation of the physiology of the host cell. It's exact role is not currently known (2008).

Descripción: MSY2 and YB-2 (MSY3,4) belong to the Y-box family of multifunctional proteins that regulate both transcription and translation (1–3). Y-box proteins interact with a wide variety of nucleic acid structures to act as transcription factors and mRNA masking proteins (1). The modular structure of Y-box proteins includes a highly conserved N-terminal cold-shock domain (CSD, equivalent to the bacterial cold-shock proteins) and four basic C-terminal domains containing arginine clusters and aromatic residues (4). MSY2 is expressed in testis and ovary where it may repress translation of parental mRNA (5,6). The gene encoding human MSY2 maps to chromosome 17p11.2-13.1 (5). YB-2 (MSY3,4 in mouse) is also known as DNA binding protein A and is highly expressed in the testis, heart and muscle (7,8). MSY2 and YB-2 bind to the consensus sequence 5'-UCCAUCA-3' contained in the Y-box element (9).

Descripción: BLK ( B lymphoid tyrosine kinase), with 505-amino acid protein (about 56 kDa), belongs to the Src non-receptor tyrosine kinases family.Different subcellular localizations of Src-family kinases may be important for the regulation of specific cellular processes such as mitogenesis, cytoskeletal organization, and membrane trafficking. Blk is expressed exclusively by B lymphocytes and it is thought to function in a signal transductory pathway specific to this lineage. B lymphoid expression of an active Blk mutant caused proliferation of B progenitor cells and enhanced responsiveness of these cells to interleukin 7. Thus, sustained activation of Blk induces responses normally associated with the pre-BCR.

Descripción: The initiation of DNA replication is a multi-step process that depends on the formation of pre-replication complexes, which trigger initiation (1). Among the proteins required for establishing these complexes are the origin recognition complex (ORC) proteins (1). ORC proteins bind specifically to origins of replication where they serve as scaffold for the assembly of additional initiation factors (1). Human ORC subunits 1-6 are expressed in the nucleus of proliferating cells and tissues, such as the testis (2). ORC1 and ORC2 are both expressed at equivalent concentrations throughout the cell cycle; however, only ORC2 remains stably bound to chromatin (3,4). ORC4 and ORC6 are also expressed constantly throughout the cell cycle (5,6). ORC2, ORC3, ORC4 and ORC5 form a core complex upon which ORC6 and ORC1 assemble (7,8). The formation of this core complex suggests that ORC proteins play a crucial role in the G1-S transition in mammalian cells (8).

Descripción: G protein-coupled receptors (GPRs or GPCRs), also known as seven transmembrane receptors, heptahelical receptors, or 7TM receptors, are members of the largest protein family and play a role in many different stimulus-response pathways. G-protein coupled receptors mediate extracellular signals into intracellular signals (G-protein activation). They respond to a great variety of signaling molecules, including hormones, neurotransmitters and other proteins and peptides. GPR proteins are integral seven-pass membrane proteins with some conserved amino acid regions. G-protein coupled receptor 56 (GPR56), also designated TM7XN1 protein, contains one GPS domain. GPR56 plays an important role in cell-cell interactions and is widely expressed, with highest levels detected in brain, heart and thyroid gland. Defects in the gene encoding for GPR56 can cause bilateral frontoparietal polymicrogyria (BFPP) which is characterized by disorganized cortical lamination.

Descripción: This MAb recognizes the 56.5 kDa (CK10); 50 kDa (CK14); 50 kDa (CK15); 48 kDa (CK16); 40 kDa (CK19) keratins of the acidic (Type I or LMW) subfamily. Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI 6.0) subfamilies. The acidic keratins have molecular weights (MW) of 56.5, 55, 51, 50, 50', 48, 46, 45, and 40 kDa. MAb AE3 recognizes the 65-67, 64, 59, 58, 56, and 52 kDa keratins of basic subfamily. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. AE1/AE3 is a broad spectrum anti pan-keratin antibody cocktail, which differentiates epithelial tumors from non-epithelial tumors e.g. squamous vs. adenocarcinoma of the lung, liver carcinoma, breast cancer, and esophageal cancer.

Descripción: The antibody recognizes the hidden determinant of β-2 microglobulin (i.e. binding to its determinant is available only when the chain is separated from the HLA heavy chain. β-2 microglobulin is a 12 kDa protein with a pI of 5.6. Serum β2 microglobulin levels are a reflection of cell turnover. Levels rise with fever, inflammation, and infection. Increased serum levels are also seen in B-cell malignancies and in renal failure and may indicate a worse prognosis for patients with early-stage Hodgkin's lymphoma. In urine, increased levels are seen in proximal renal tubular disease as well as renal transplant rejection. β2 microglobulin levels can rise either because its rate of synthesis has increased (e.g. in AIDS, malignant monoclonal plasma cell dyscrasia, solid tumours and autoimmune disease) or because of impaired renal filtration (e.g. due to renal insufficiency, graft rejection or nephrotoxicity induced by post-transplantation immunosuppressive therapy).
