Descripción: Protein arginine deiminase 6 (PAD6) is a homodimeric guanidine-modifying enzyme belonging to the amidinotransferase superfamily. It is a calcium-dependent enzyme that catalyzes the post-translational modification of target proteins by converting arginine to citrulline. PAD6 is expressed in mammalian oocytes, sperm cells, and early embryos. In mammalian oocytes and early embryo cytoplasm, its expression is localized to cytoskeletal sheets, dynamic structures containing various keratins, which are major targets for citrullination. PAD6-/- oocytes exhibit reduced microtubule acetylation and defective organelle positioning and redistribution, suggesting a role for PAD6 in regulating microtubule-mediated organelle movement and positioning. PAD6-/- female, but not male, mice are infertile due to a reduction of de novo protein synthesis, cytoskeletal sheet formation, and ribosomal RNA which induces arrest of zygote development at the two-cell stage. PAD6 is regulated by newborn ovary homeobox (Nobox), as its promoter contains a Nobox DNA-binding element (NBE) and expression and activity of PAD6 are decreased in Nobox-/- mouse ovaries.5 In human females, homozygous nonsense mutations and compound-heterozygous mutations in PAD6 induce early embryonic arrest following in vitro fertilization (IVF) or intracytoplasmic sperm injection (ICSI).

Descripción: Coenzyme A is an essential cofactor in enzymatic acetyl transfer reactions. The principal biologically active forms of pantothenic acid are coenzyme A (CoA) and acyl carrier protein (ACP). In CoA, the business center of the molecule is the pantothenic acid metabolite 4'-phosphopantetheine. Coenzyme A is comprised of 4'-phosphopantetheine linked by an anhydride bond to the nucloetide adenosine 5'-monophosphate. 4'-Phosphopantetheine itself is comprised of pantothenic acid linked at one end, via an amide bond, to beta-mercaptoethylamine, derived from L-cysteine, and at the other end to a phosphate group. The sulfhydryl group of 4'-phosphopantetheine, which is the business end of the coenzyme, forms thioesters with acyl groups producing acyl-CoA derivatives, including acetyl-CoA.
Coenzyme A facilitates removal of lipid peroxides by increasing mobilization of fatty acids, and promote repair of plasma membranes by activating phospholipid synthesis.

Descripción: Histone methyltransferases (HMTases) selectively methylate evolutionarily conserved arginine or lysine residues, primarily in the N-terminal tails of histones H3 and H4. Signal transduction pathways affecting the N-terminal tails of histones lead to a number of post-translational modifications including acetylation, phosphorylation, poly(ADP-ribosylation), ubiquitination and methylation. These modifications play critical roles in regulating chromatin structure and gene expression. Set7/9 is a histone specific HMTase that methylates histone H3 lysine 4. Set7/9 transfers methyl groups to lysine 4 of histone H3 in complex with S-adenosyl-L-methionine. In yeast, H4-K20 methylation does not have any apparent role in the regulation of gene expression or heterochromatin function; rather it appears to play a role in DNA damage response. Loss of Set9 activity or mutation of H4-K20 markedly impairs yeast cell survival after genotoxic challenge and compromises the ability of cells to maintain checkpoint mediated cell cycle arrest. Genetic experiments link Set9 to Crb2, a homolog of the mammalian checkpoint protein 53BP1, and the enzyme is required for Crb2 localization to sites of DNA damage.
