El reactivo, una solución de fase única que consta de fenol e isotiocianato de guanidina, es una modificación del método de aislamiento de RNA de un paso desarrollado por Chomczynski y Sacchi. La muestra se homogeniza y lisa en reactivo RNA-Solv®, que mantiene la integridad del RNA a la vez que altera y desnaturaliza las RNAsas endógenas y otros componentes celulares. La extracción del lisado con cloroformo desnaturaliza aún más las proteínas y separa la mezcla en una fase orgánica y una fase acuosa. El RNA permanece exclusivamente en la fase acuosa y, posteriormente, se recupera mediante isopropanol.
